This variant lies in the DSP gene (transcript NM_004415.4) at the canonical splice donor site of the intron immediately after coding-DNA position 939, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 7 of the DSP gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with clinical features of DSP-related conditions (PMID: 10594734, 20864495; internal data). ClinVar contains an entry for this variant (Variation ID: 1076116). Studies have shown that disruption of this splice site results in intron 7 inclusion, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 10594734).
