Pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_23625315)_(23625422_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 11 of the PALB2 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in PALB2 are known to be pathogenic (PMID: 17200668, 17200671, 17200672, 24136930, 25099575). A similar copy number variant has been observed in individual(s) with breast and ovarian cancer, cervical and breast cancer, and/or prostate cancer (PMID: 24982446, 26681312, 27433846). For these reasons, this variant has been classified as Pathogenic.
