Pathogenic for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000011.9:g.(?_47363401)_47367757del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exon(s) 13-18 (c.1090+1_1790+141del) of the MYBPC3 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with MYBPC3-related conditions. This variant disrupts the p.Gly531Arg amino acid residue in MYBPC3. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 18533079, 21750094, 27600940, 28356264, 23508784). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Loss-of-function variants in MYBPC3 are known to be pathogenic (PMID: 19574547). For these reasons, this variant has been classified as Pathogenic.
