Pathogenic for Fanconi anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_89842140)_(89866056_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 9-21 of the FANCA gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with FANCA-related disease. However, smaller gross deletions encompassed within this deleted region have been observed in individuals affected with¬†Fanconi anemia (PMID:¬†16084127,¬†16015582). Loss-of-function variants in FANCA are known to be pathogenic (PMID: 19367192). For these reasons, this variant has been classified as Pathogenic.
