NC_000003.11:g.(?_167422610)_(167437965_?)del was classified as Pathogenic for Cerebral cavernous malformation 3 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 2-3 of the PDCD10 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 3 of the PDCD10 gene. This is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in PDCD10 are known to be pathogenic (PMID: 15543491, 18300272, 23801932). Similar deletions have been observed in individual(s) with diagnosis of, or clinical features of, cerebral cavernous malformations (PMID: 23595507, 23801932).
