Pathogenic for Fanconi anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_89804999)_(89825128_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 30-43 of the FANCA gene. The 5' boundary is likely confined to intron 29. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. Similar deletions of exons 30-43 have been observed in individual(s) with Fanconi anemia (PMID: 29098742). The region of the FANCA gene that includes exon(s) 37-43 has been determined to be clinically significant (PMID: 23613520, Invitae). Therefore, deletions that encompass that region are likely to disrupt protein function and cause disease. For these reasons, this variant has been classified as Pathogenic.
