This variant lies in the COL2A1 gene (transcript NM_001844.5) at coding-DNA position 4293, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 1431 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the COL2A1 gene (p.Tyr1431*). While this is not anticipated to result in nonsense mediated decay, it is expected to delete the last 57 amino acids of the COL2A1 protein. This variant has been observed to be de novo in an individual affected with clinical features of COL2A1-related disease (Invitae). Other truncations (such as p.C1438*, p.K1447*, p.Leu1434*) that lie downstream of this variant have been reported in individuals affected with COL2A1-related disease (PMID: 15316962, 21356074, 26443184).
