Pathogenic for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma — the classification assigned by All of Us Research Program, National Institutes of Health to NM_004415.4(DSP):c.3739C>T (p.Arg1247Ter), citing ACMG Guidelines, 2015: This variant changes 1 nucleotide in exon 23/24 of the DSP gene, creating a premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. This variant has been reported in an individual affected with dilated cardiomyopathy (PMID: 31983221). This variant has been identified in 1/31380 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Loss of DSP function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
