Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.11:g.(?_10393732)_(10439975_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in TFAP2A are known to be pathogenic (PMID: 8622766, 20150232, 21204207, 21539471, 21728810). Experimental studies have shown that this deletion could lead to eye and craniofacial defects in a model organism (PMID: 19685247). A similar deletion of the TFAP2A gene has been reported in a family affected with branchio-oculo-facial syndrome¬†(PMID: 19685247). A gross deletion of the genomic region encompassing the full coding sequence of the TFAP2A gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes.
