Pathogenic for FOXG1 disorder — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005249.5(FOXG1):c.1410del (p.Leu471fs), citing Invitae Variant Classification Sherloc (09022015): This variant disrupts the C-terminus of the FOXG1 protein. Other variant(s) that disrupt this region (p.Ser472Ilefs*15) have been determined to be pathogenic (Invitae). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic. This variant has been observed in individual(s) with FOXG1-related conditions (Invitae). This sequence change results in a premature translational stop signal in the FOXG1 gene (p.Leu471Cysfs*17). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 19 amino acids of the FOXG1 protein. This variant is not present in population databases (ExAC no frequency).
