Pathogenic for Nemaline myopathy 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001164508.2(NEB):c.18981C>A (p.Tyr6327Ter), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 1074050). This premature translational stop signal has been observed in individual(s) with clinical features of congenital nemaline myopathy (PMID: 29274205). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr6327*) in the NEB gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NEB are known to be pathogenic (PMID: 25205138). For these reasons, this variant has been classified as Pathogenic.
