Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_29422322)_(29701178_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the NF1 gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Similar deletions of NF1 including larger contiguous gene deletions have been observed in individual(s) with neurofibromatosis type 1 (PMID: 10631140, 10587576, 25541118, 22837079, 25480383) and in the mosaic state in individual(s) without clinical features of neurofibromatosis type 1 (PMID: 23532973). Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). For these reasons, this variant has been classified as Pathogenic.
