Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000497.4(CYP11B1):c.421C>T (p.Arg141Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Arg141*) in the CYP11B1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CYP11B1 are known to be pathogenic (PMID: 8506298, 26476331). This variant is present in population databases (rs775479837, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with 11Œ≤-hydroxylase deficiency (PMID: 27316665). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
