Pathogenic for Landau-Kleffner syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_9916101)_(9923529_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exons 10 and 11 of the GRIN2A gene. While the exact position of this variant cannot be determined from this data, sub-genic copy number gains are generally in tandem (PMID: 25640679) and may result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in GRIN2A are known to be pathogenic (PMID: 23933819, 23933820). This variant has been observed in individual(s) with clinical features of GRIN2A-related conditions (Invitae). In at least one individual the variant was observed to be de novo.
