Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_41256129)_(41258568_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 4-6 of the BRCA1 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Similar deletion of exons 4-6 has been observed in individuals and families affected with breast and/or ovarian cancer (PMID: 18546071, 16715518, 16772120, Invitae). Deletion of exons 4-6 is also known as deletion of exons 5-7 in the literature. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). For these reasons, this variant has been classified as Pathogenic.
