Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in RB1 are known to be pathogenic (PMID: 17096365). Disruption of this splice site has been observed in individuals with bilateral retinoblastoma (PMID: 18181215, 12402348, 15884040). This variant is also known as IVS2+1G>T in the literature.
