Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042492.3(NF1):c.1261-1G>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects an acceptor splice site in intron 11 of the NF1 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in the loss of 8 amino acid residue(s), but is expected to preserve the integrity of the reading-frame. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with neurofibromatosis type 1 (PMID: 18041031, 24232412, 29618358, 30530636; Invitae). ClinVar contains an entry for this variant (Variation ID: 1073144). Studies have shown that disruption of this splice site results in the activation of a cryptic splice site in exon 12 (Invitae). For these reasons, this variant has been classified as Pathogenic.
