This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 606, where T is replaced by G; at the protein level this means replaces cysteine at residue 202 with tryptophan — a missense variant. Submitter rationale: GLA c.606T>G is a missense variant that changes the amino acid at residue 202 from Cysteine to Tryptophan. This variant has been observed in at least one proband affected with Fabry disease (PMID:10208848;8012363;25511234;25974833;23566439). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:27657681). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify GLA c.606T>G as a pathogenic variant.
