Pathogenic for Combined immunodeficiency due to DOCK8 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.11:g.(?_325651)_(386446_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 8-23 of the DOCK8 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in DOCK8 are known to be pathogenic (PMID: 14722525, 19776401). For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with DOCK8-related conditions.
