Loss-of-function variants in USH1C are known to be pathogenic (PMID: 10973247, 17407589, 20301442, 21203349). This variant has not been reported in the literature in individuals with USH1C-related conditions. This variant is an out-of-frame deletion of the genomic region encompassing exons 7-15 of the USH1C gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This deletion may extend further upstream beyond exon 7, however, this cannot be determined by this assay because del/dup assessment is not offered for exons 5-6 of the USH1C gene.
