This variant disrupts the C-terminus of the MLH1 protein. Other variant(s) that disrupt this region (p.Lys751Serfs*3) have been determined to be pathogenic (PMID: 24802709, 8797773, 27295708, 18566915, 18931482, 12799449, 16338176, 20533529). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. This variant has not been reported in the literature in individuals with MLH1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change results in a frameshift in the MLH1 gene (p.Met682Ilefs*99). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 75 amino acids of the MLH1 protein and extend the protein by an additional 23 amino acids.
