This variant disrupts the p.Cys140 amino acid residue in LMX1B. Other variant(s) that disrupt this residue have been observed in individuals with LMX1B-related conditions (PMID: 11668639, 20531206), which suggests that this may be a clinically significant amino acid residue. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has been observed in individual(s) with nail-patella syndrome (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (ExAC no frequency). This sequence change replaces cysteine with phenylalanine at codon 140 of the LMX1B protein (p.Cys140Phe). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and phenylalanine.
