This variant lies in the LAMA2 gene (transcript NM_000426.4) at coding-DNA position 1824, where T is replaced by G; at the protein level this means converts the codon for tyrosine at residue 608 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This nonsense change has been observed in individual(s) with congenital muscular dystrophy (PMID: 30055037). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Tyr608*) in the LAMA2 gene. It is expected to result in an absent or disrupted protein product.
