This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 131, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 44 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: GLA p.Trp44Ter (c.131G>A) is a nonsense variant that introduces a premature stop codon at amino acid position 44, creating a truncated protein that is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:9105656;30386727;23818648;27585509;2171331). Functional studies have been reported; however, the significance of the findings remain unclear and/or they were performed in patient cells (PMID:23818648;27585509). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Trp44Ter (c.131G>A) as a pathogenic variant.
