Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.11:g.(?_94485944)_(94538110_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 2-9 of the ROR2 gene. The 5' boundary is likely confined to intron 1. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. This variant has been observed in individual(s) with clinical features of Robinow syndrome (Invitae). The region of the ROR2 gene that includes exons 6-7 has been determined to be clinically significant (PMID: 18831060). Therefore, deletions that encompass that region are likely to disrupt protein function and cause disease. For these reasons, this variant has been classified as Pathogenic.
