NM_005802.5(TOPORS):c.1882C>T (p.Arg628Ter) was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. A different truncation (p.Arg847*) that lies downstream of this variant has been determined to be pathogenic (PMID: 22581970, 28453362, 26155838). This suggests that deletion of this region of the TOPORS protein is causative of disease. This variant has not been reported in the literature in individuals with TOPORS-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the TOPORS gene (p.Arg628*). While this is not anticipated to result in nonsense mediated decay, it is expected to delete the last 418 amino acids of the TOPORS protein.
