Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_41226328)_(41234612_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 12-14 of the BRCA1 gene. This creates a premature translational stop signal and is expected to result in an absent or disrupted protein product. Deletion of exons 12-14 has been observed in individuals affected with breast/ovarian cancer (PMID: 117470134, 20617377, 18431737, 19669600). Deletion of exons 12-14 is also known as a deletion of exons 13-15 in the literature. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). For these reasons, this variant has been classified as Pathogenic.
