Pathogenic for Werner syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000553.6(WRN):c.3310-1_3369dup, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WRN gene (transcript NM_000553.6) at the canonical splice acceptor site of the intron immediately before coding-DNA position 3310 through coding-DNA position 3369, duplicating this region. Submitter rationale: Loss-of-function variants in WRN are known to be pathogenic (PMID: 16673358). For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with WRN-related conditions. This sequence change creates a premature translational stop signal (p.Ile1124Valfs*2) in the WRN gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency).
