Pathogenic for Short-rib thoracic dysplasia 14 with polydactyly; Joubert syndrome 23 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001329943.3(KIAA0586):c.4495+3755del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the KIAA0586 gene (transcript NM_001329943.3) at 3755 bases into the intron immediately after coding-DNA position 4495, deleting one base. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. ClinVar contains an entry for this variant (Variation ID: 1070838). This variant has not been reported in the literature in individuals affected with KIAA0586-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Val1555Tyrfs*3) in the KIAA0586 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in KIAA0586 are known to be pathogenic (PMID: 26096313, 26166481, 26386044).
