This variant disrupts the p.Asp268 amino acid residue in ADSL. Other variant(s) that disrupt this residue have been observed in individuals with ADSL-related conditions (PMID: 18524658), which suggests that this may be a clinically significant amino acid residue. This variant has been reported to affect ADSL protein function (PMID: 10888601, 20127976). This variant has been observed in individual(s) with adenylosuccinate lyase deficiency (PMID: 10888601, 20127976, 23055421, 29302074, Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs746501563, ExAC 0.01%). This sequence change replaces aspartic acid with asparagine at codon 268 of the ADSL protein (p.Asp268Asn). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and asparagine.
