This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. A similar copy number variant has been observed in individual(s) with Bardet-Biedl syndrome (PMID: 27486776, Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in BBS1 are known to be pathogenic (PMID: 12118255).
