This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 479 of the CYP27A1 protein (p.Arg479Ser). This missense change has been observed in individual(s) with cerebrotendinous xanthomatosis (PMID: 29095540). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Arg479 amino acid residue in CYP27A1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 2019602, 21073839, 24584636). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Experimental studies have shown that this missense change affects CYP27A1 function (PMID: 17444890).
