Pathogenic for Diffuse cerebral and cerebellar atrophy - intractable seizures - progressive microcephaly syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005051.3(QARS1):c.1485dup (p.Lys496Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Lys496*) in the QARS gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in QARS are known to be pathogenic (PMID: 24656866, 25471517). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with early-onset epileptic encephalopathy (PMID: 25471517). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1069375). For these reasons, this variant has been classified as Pathogenic.
