Pathogenic for Familial focal epilepsy with variable foci — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001242896.3(DEPDC5):c.2548dup (p.Asp850fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DEPDC5 gene (transcript NM_001242896.3) at coding-DNA position 2548, duplicating one base; at the protein level this means shifts the reading frame starting at aspartic acid residue 850, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Asp850Glyfs*19) in the DEPDC5 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in DEPDC5 are known to be pathogenic (PMID: 23542697, 23542701). This variant has not been reported in the literature in individuals with DEPDC5-related disease.
