Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_16248465)_(16259810_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 23-29 of the ABCC6 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in ABCC6 are known to be pathogenic (PMID: 11536079, 17617515). A similar copy number variant has been observed in individual(s) with pseudoxanthoma elasticum and generalized arterial calcification of infancy (PMID: 11179012, 11536079, 17724214, 22209248). It has also been observed to segregate with disease in related individuals. For these reasons, this variant has been classified as Pathogenic.
