Pathogenic for 6-Pyruvoyl-tetrahydrobiopterin synthase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000011.9:g.(?_112096088)_(112105696_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the PTS gene has been identified. Loss-of-function variants in PTS are known to be pathogenic (PMID: 3297709, 16917893). The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. Isolated whole-gene deletions of PTS have not been reported in the literature. However, larger copy number events that include this gene have been reported (PMID: 10874306). For these reasons, this variant has been classified as Pathogenic.
