Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030632.3(ASXL3):c.1465G>T (p.Glu489Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu489*) in the ASXL3 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ASXL3 cause disease. This variant has been observed in individual(s) with clinical features of Bainbridge-Ropers syndrome (Invitae). In at least one individual the variant was observed to be de novo.
