Submitter rationale: This sequence change replaces valine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 184 of the TSC2 protein (p.Val184Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of tuberous sclerosis complex (PMID: 33448881, 35723786).
