Glycine residues within the Gly-Xaa-Yaa repeats of the triple helix domain are required for the structure and stability of fibrillar collagens (PMID: 7695699, 8218237, 19344236). This suggests that a deletion that includes this region may also be pathogenic. This variant has not been reported in the literature in individuals affected with COL11A1-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 2-67 of the COL11A1 gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
