Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 251380 control chromosomes. c.238C>A has been observed in individuals affected with Autosomal Recessive Non-Syndromic Hearing Loss (e.g. Kalay_2005, Gruber_2016, Abbaspour Rodbaneh_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. However, a different variant affecting the same codon has been classified as pathogenic by our lab (c.239A>C, p.Gln80Pro), supporting the critical relevance of codon 80 to GJB2 protein function.
