Pathogenic for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002047.4(GARS1):c.1007C>G (p.Pro336Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 336 of the GARS protein (p.Pro336Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with congenital hypomyelinating polyneuropathy (internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1067545). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GARS protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects GARS function (PMID: 35332613). For these reasons, this variant has been classified as Pathogenic.
