Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003108.4(SOX11):c.168C>G (p.Phe56Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 56 of the SOX11 protein (p.Phe56Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of Coffin-Siris syndrome (PMID: 35341651; internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1067424). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt SOX11 protein function with a positive predictive value of 95%.
