This variant lies in the LAMA2 gene (transcript NM_000426.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1884, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: LAMA2 c.1884+2T>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of LAMA2 function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250706 control chromosomes.
