NM_005866.4(SIGMAR1):c.492G>A (p.Trp164Ter) was classified as Pathogenic for Amyotrophic lateral sclerosis type 16; Autosomal recessive distal spinal muscular atrophy 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Trp164*) in the SIGMAR1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 60 amino acid(s) of the SIGMAR1 protein. For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the SIGMAR1 protein in which other variant(s) (p.Asn167Ile) have been determined to be pathogenic (PMID: 31511340). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 1067324). This variant has not been reported in the literature in individuals affected with SIGMAR1-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.0009%).
