Pathogenic for Mucopolysaccharidosis type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000203.5(IDUA):c.1858G>T (p.Val620Phe), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 1067173). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt IDUA protein function. For these reasons, this variant has been classified as Pathogenic. This sequence change replaces valine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 620 of the IDUA protein (p.Val620Phe). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with IDUA-related conditions (PMID: 19396826). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
