This variant lies in the CALM2 gene (transcript NM_001743.6) at coding-DNA position 397, where G is replaced by A; at the protein level this means replaces glycine at residue 133 with serine — a missense variant. Submitter rationale: This sequence change replaces glycine with serine at codon 133 of the CALM2 protein (p.Gly133Ser). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and serine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with clinical features of long QT syndrome (Invitae). In at least one individual the variant was observed to be de novo. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55").
