This variant lies in the GBE1 gene (transcript NM_000158.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1804, where A is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: GBE1 c.1804-2A>T is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4.3e-06 in 229908 control chromosomes.
