NM_004484.4(GPC3):c.1569del (p.Ala524fs) was classified as Pathogenic for Wilms tumor 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the GPC3 protein in which other variant(s) (p.G556R) have been determined to be pathogenic (PMID: 19215053, 29637653). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 1067035). This variant has not been reported in the literature in individuals affected with GPC3-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ala524Glnfs*26) in the GPC3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 57 amino acid(s) of the GPC3 protein.
