This variant would be expected to be in-frame, preserving the integrity of the reading frame. A similar copy number variant has been observed in individual(s) with hypophosphatasia (PMID: 33549410). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant disrupts a region of the ALPL protein in which other variant(s) (p.Asp294Ala) have been determined to be pathogenic (PMID: 1409720, 10839996, 15694177, 25731960). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
