Likely pathogenic for Chromosome 2q32-q33 deletion syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.11:g.(?_200320572)_(200320780_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exon 3 of the SATB2 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 3 of the SATB2 gene. As the precise location of this event is unknown, it may be in tandem or it may be located elsewhere in the genome. A similar gain of exon 3 has been observed in individual(s) with SATB2-related conditions (PMID: 25118029). In at least one individual the variant was observed to be de novo. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
